1) Conduct full factorial field experiment (n = 4) over three growing seasons in which canopy temperatures are elevated 6 deg C above ambient for 5 days with and without elevated [CO2] (600 ppm) on soybean and soybean over expressing SBPase. Heat waves will be applied on separate plots during 3 different developmental states (V4, R2, R5). 2) Quantify by gas exchange and chlorophyll fluorescence analysis the extent of RuBP regeneration vs. Rubisco limitation and determine extent of acclimation for all treatments. 3) Quantify by western blotting the amount of enzymes implicated by control flux analysis (Rubisco, Rubisco activase, cytochrome b6f, SPBase, transketolase, FBPase and aldolase). 4) Quantify treatment impacts on photosynthetic energy conversion efficiency, biomass accumulation, seed yield and harvest index.
FACE technology coupled with and IR canopy heaters currently in operation at SoyFACE research site (http://www.igb.illinois.edu/soyface/) will be used to impose the treatments on soybean and soybean already transformed to over express SBPase.
