1. To develop and assess a genetic mapping population derived from Fiskeby soybean to identify molecular markers associated with genes for resistance to iron deficiency chlorosis, drought, salt stress, aluminum stress, and ozone stress. 2. Apply these molecular markers to identify and develop stress resistant cultivars.
Fiskeby soybean ancestors have been identified as sources of resistance to multiple stress factors including drought, iron deficiency chlorosis, salt, and ozone air pollution. A cross made between Fiskeby III and Mandarin-Ottawa (known to be sensitive to these stress factors) has been used to develop a mapping population of 240 recombinant inbred lines (RILs). The RILs have been assayed for single nucleotide polymorphisms (SNP) to identify DNA markers for mapping stress resistant genes. The RILs will be assessed for response to drought, iron deficiency chlorosis, salt tolerance, aluminum tolerance, and ozone tolerance. Injury scores and DNA marker results will be related through QTL analysis. Specific DNA markers associated with stress resistance genes from Fiskeby III will be used to identify modern cultivars derived from the Fiskeby pedigree that will be tested for stress tolerance. Fiskeby lines will be crossed with cultivars for different soybean maturity groups and the DNA makers used to select breeding lines carrying Fiskeby stress resistance genes.
