If the core represents the basis for the variation found in the peanut germplasm in the US then a more complete evaluation and characterization of the collection should be conducted. We propose to evaluate the entire core collection over a two year period, assessing phentypic (yield and grade), biochemical, and genetic traits. The characterization study will be conducted under field conditions in Citra, FL. We will utilize an augmented design over two years, where lines within the core collection will be compared to current production lines.
We propose to regenerate the core collection over a two year period in Citra, FL. The core will be compared to current production lines. Yield and grade data will be collected on each accession. After harvesting biochemical data will be collected from the regenerated seeds. We will use a gas chromatograph to collect fatty acid data. A near infrared machine will be used to evaluate protein and sugar content. DNA will be extracted from each accession by following the instructions from an Omega Bio Tek Kit. All DNA samples will be quantified and genotyped with SSR markers.
