Examine the spread of wood-canker pathogens in grapevine wood.

This research is focused on developing an infection assay for wood-canker pathogens, and is one component of a related project on early detection of trunk diseases of grape. The long-term goal of this research and the related project is to identify a 'molecular signature' of gene expression that is unique to the early stage of infection, and then to develop a PCR-based method for detecting the molecular signature from asymptomatic leaves. The first step, which is the focus of this research, is to define the early stage of infection, in terms of colonization beyond the inoculation site and/or plant anatomical responses to infection (e.g., xylem occlusions). The second step, which is the focus of the related project mentioned above, is to identify candidate genes in asymptomatic grape leaves, as detected by RNA-seq. and confirmed by qRT-PCR, the expression of which corresponds with the early stage of infection in the woody stem.
