The first phase of this study of the effects of probiotics on children with Cystic Fibrosis (CF) is to evaluate the long term effects of probiotic Lactobacillus rhamnossus (LGG) on the host transcriptome of healthy adults during a longitudinal six month intervention study with one and six month follow-up evaluation after termination of intervention. Effect of probiotic intervention will be assessed by analysis of differentially expressed genes (DEG) during and after probiotic intervention. DEG data analysis will be associated with metabolic and immunological pathways to predict effect of probiotic intervention on metabolic and immunological function and safety.

Cooperator will provide blood samples collected from healthy subjects at multiple samples times (baseline, day 28, months 3,6,7,12) from a randomized control study monitored by the Cooperator’s clinical team. After shipment of samples to Beltsville, nucleic acid will be isolated and processed in ARS laboratories. Host transcriptome analysis will be done by deep-sequencing analysis (Illumina platform) using at least 250 ng of globin-treated RNA extracted from whole blood. DEG (differentially expressed gene) analysis will be done with appropriate software to predict changes in host response.
