1a. Objectives (from AD-416):
To design and optimize dsRNA inhibitor of apoptosis (IAP) constructs for incorporation into an attractive sugar bait product with enhanced uptake for control of mosquitoes and sand flies.
1b. Approach (from AD-416):
1. DsRNA constructs will be optimized for enhanced activity in silencing the targeted genes in mosquitoes and sand flies. 2. The environmental fate of dsRNA in various sugar baits will be evaluated to determine stability profiles over time. This information is crucial for determining the effective life of deployed sugar baited traps. 3. Optimized dsRNA constructs and formulations will be evaluated in laboratory and field trials against Ae. aegypti.
3. Progress Report:
This work directly relates to inhouse objective 1. Discover and evaluate new toxicants and biological control agents for control of biting Nematocera, and 3. Conceive and test new methods of managing vector and pest populations through the use of behavior-altering chemicals, including repellents, attractants, and inhibitors. This report covers the first year of a project to investigate gene silencing as method for mosquito control. Funding has been provided by the AFPMB for a molecular biologist and technician to conduct the research and these positions have been filled as of August 2013. Studies have been initiated to determine the stability of dsRNA constructs in various sugar bait formulations. Milligram quantities of IAP and other constructs have been obtained to conduct sugar feeding assays against Aedes aegypti adults to determine stability and effectiveness of these constructs in silencing the targeted genes. The most effective constructs will be selected for additional formulation and evaluation.
