1a. Objectives (from AD-416):
The objectives are to develop mutants of Streptomyces that are deficient in expression of three family-12 endoglucanases. Develop mutants of additional pathogenicity-related genes and to train scientists in Streptomyces mutagenesis.
1b. Approach (from AD-416):
Individual genes or gene fragments will be cloned and transferred to appropriate vectors for use in mutagenesis experiments. Tests will be conducted to determine the effect of mutagenesis on development of Streptomyces-induced common scab disease.
3. Progress Report:
Microbes such as Streptomyces scabies can cause disease on crop plants. Understanding how they cause disease would aid in determining methods for limiting or preventing crop losses. Numerous genes have been identified in S. scabies that may contribute to causing disease. Three of these genes, affecting plant cell walls, have been targeted for disruption, and the mutated isolates are ready for testing.
