1b. Approach (from AD-416):
a. Allelism tests of downy mildew resistance loci in cucumber: inbred lines carrying multi-source downy mildew resistance genes will be crossed and segregation of resistance will be evaluated in multi-year and location field and greenhouse trials. b. Molecular mapping of downy mildew resistance: Multiple mapped populations will be developed and phenotypic data of downy mildew resistance will be collected. Genomic DNAs of segregation populations and parental lines will be used for molecular marker analysis to identify markers for downy mildew resistance major QTL. c. The outreach program will enhance communication with various segments of the industry, and educate the public on the need for sound scientific practices to resolve biological problems. The outreach target groups will include the cucumber growers, the private seed companies, cucumber scientific community, the youth, and the general public.
3. Progress Report:
This project was renumbered from 3655-21000-048-39A to 3655-21000-062-13A. For quantitative trait loci (QTL) mapping of downy mildew (DM) resistance, we have developed several mapping populations including a recombinant inbred line (RIL) population from plant introduction line (PI) 197088 × Coolgreen and PI 197088 x Poinsett 76. Several F2 populations among old and new resistance sources are being developed for study of inheritance and allelism tests. These populations are growing in the field for screening of DM research upon artificial inoculation this growing season. We attended a number of stakeholders’ meetings or professional meetings, gave presentations and organized workshops aiming to enhance communication with various segments of the industry and to educate the public on the need for sound scientific practices to resolve biological problems. This work relates to objective 1 by determining the genetic basic of disease resistance and cucurbits and objective 2 by utilizing current biotechnology to discover and evaluate genetic variation and to map agriculturally important traits in cucurbits.
