1a. Objectives (from AD-416):
We propose to develop, characterize, and advance breeding populations created to improve germination and emergence in low phytate soybeans, using two sources of the low phytate trait, CX1834-derived lines and Mips1 mutants.

1b. Approach (from AD-416):
We will develop breeding methodologies for LP (low phytate) germplasm in tandem with strategic handling of LP breeding populations for selection of improved G&E. Genetic improvement of G&E in LP germplasm should be possible through development of effective breeding methodologies, germplasm characterization, and selection of appropriate parents.

3. Progress Report:
This project is related to Objective 2 of the in-house project: To discover novel genes/alleles in soybean for seed composition, determine their inheritance, determine genomic location, transfer to adapted germplasm, and release. In 2011, approximately 42 populations were grown Blacksburg, VA that combined the low phyate trait with better agronomic performance including crosses to lines that may improve field emergence. The populations were in the F2 generation and three 10' rows per population (pop) were planted. The first row of each pop contained 60 seeds and those rows were assayed for emergence with stand counts. Percent emergence and visual observations were used to select 26 populations for marker-assisted selection. DNA was isolated from individual F2 plants in each population; 320 plants were assayed in Lpa pops and 80 in Mips1 pops. Thus far, approximately 6,000 F2 plants have been assayed with single nucleotide polymorphism (SNP) markers.
