1a. Objectives (from AD-416)
To develop high-yielding low phytic acid basmati rice in Pakistan.

1b. Approach (from AD-416)
Alleles which impart the low phytic acid trait in rice will be isolated from mutagenized populations of Pakistani rices. Using conventional methods, these will be used to breed high yielding low phytic acid rices for production in Pakistan.

3. Progress Report
This work contributes to National Program 301, Component 2, Crop Informatics, Genomics and Genetic Analyses, Problem 2C, Genetic Analyses and Mapping of Important Traits and relates to objective 1 of the parent project. The traits in question concern the nutritional quality of seed crops. This parent project focuses on the genetics of plant and seed phosphorus and phytic acid. Both the total amount of phosphorus and its chemistry are critically important to the end-use quality of grain and legume crops. Several lines of work were begun in Pakistan and are underway in the first year of this project. First, a survey of Pakistani Basmati rice cultivars and germplasm for the phytic acid seed trait is underway, using simple lab tests for seed phosphorus composition and using molecular markers for known genes important to seed phytic acid. Second, populations of randomly induced mutations in Basmati rice lines are being grown out for subsequent screening, using methods developed by the USDA-ARS, for novel mutations that produce the low phytic acid trait. Third, in a prior project the original rice low phytic acid alleles isolated by the USDA-ARS were crossed into locally-adapted Pakistani Basmati rice cultivars. In the first year of this new project, this germplasm will be used in a plant breeding program to develop first generation Pakistani low phytic acid Basmati rices. These new cultivars will then be the subject of field evaluations of agronomic performance, and of laboratory analyses of grain quality. Communication between cooperators on this project is via the internet and e-mail communication.
