1a. Objectives (from AD-416):
Conduct a two year multi-location association mapping study to develop marker-trait associations to facilitate the development of new oat varieties with superior agronomics and milling quality.
1b. Approach (from AD-416):
Single row plots for all 685 association lines will be grown in an augmented design. Agronomic traits evaluated prior to harvest will include; 1) heading date, 2) plant height, 3) lodging, and 4) maturity. Four hundred grams of seed will be harvested and sent to Aberdeen, ID for quality analysis including: 1) groat percentage, 2) percentage plump kernels, 3) 1000 kernel weight, 4) groat plumpness, 5) groat color, and 6) percentage broken groats using standardized methods. All data will be collected and analyzed for marker trait associations. Seed will also be tested for tocopherol, total dietary fiber, beta glucan, and avenanthramides. Documents Grant with NDSU.
3. Progress Report:
This progress applies directly to Sub-objective-3B: “Develop oat cultivars combining the enhanced levels of disease resistance from southern U.S. with the superior yield and quality of ARS-Aberdeen lines” of the parent project. As part of the phenotyping for the Agronomy and Quality Testing of the Oat SNP Development and Identification of Loci, 480 spring lines, 129 CORE lines and 170 winter lines were grown in 4-row plots 1.8 m long with 30 cm row centers at NDSU Casselton, North Dakota, research site. Very wet soil conditions delayed planting until May 26, but emergence was uniform after planting. Higher than normal rainfall continued throughout the season, but little flooding injury occurred. Heading dates were recorded as days after May 31 when 50% of panicles were fully emerged from the flag leaf whorl. Only a small number winter lines did not head due to lack of vernalization while some winter lines headed very late and produced little seed. Early in the season, some lines exhibited leaf chlorosis that was noted on data sheets as CHL. Barley yellow dwarf virus (BYDV) infection was severe in some lines and was noted as BYD on the data sheet. Heavy crown rust infection occurred on susceptible lines and produces a large effect on performance of susceptible lines. Crown rust severity notes were included in the data set and sent Texas A&M for purposes of crown rust resistance association mapping.Lodging and plant height notes were recorded and included in the data set. The center two rows of each plot were harvested using a two-row binder and threshed with a Vogel thresher. Seed from all plots that produced seed was cleaned using a Seed-Boy cleaner and proportion of kernels passing through a 5/64” sieve was determined. Two hundred g seed samples of each plot that produced seed was sent to the USDA-ARS collaborators at Aberdeen, Idaho. Data obtained from the trials was sent to cooperators at Aberdeen, Idaho, for analysis.
