1a. Objectives (from AD-416):
1) Develop genetic crosses of selected isolates of P. graminis, and P. striiformis; 2) Make collections of rust fungi in Israel; 3) Establish and monitor field trap plots for wheat rust in Israel; and 4) characterize spore types and biology of selected rust fungi.
1b. Approach (from AD-416):
Telia of selected isolates will be used to infect appropriate alternate host and genetic crosses will be made from resulting pycnia. Pure lines resulting from the crosses will be developed and characterized on selected host plants. Collections of rust fungi will be made from selected sites in Israel. Biological material will be produced and collected from infection of appropriate hosts. Experiments will be performed to test selected inhibitors on early steps in mating. Spore morphology and life-cycle biology will be characterized for selected rust samples.
3. Progress Report:
Trap nurseries were established at three locations in Israel and monitored for wheat stem rust from 2010-2013. P. graminis f. sp tritici collections were purified and characterized on standard wheat stem rust differentials for samples from 2010-2012. Analysis of race phenotypes showed that P. graminis f. sp. tritici population shifts occurred yearly indicating that the P. graminis f. sp. tritici population in Israel is part of larger regional population. Selected isolates were increased and submitted for genotyping as part of a global Pgt project. Methods were developed for producing biological materials for making genetic crosses of P. graminis f. sp. tritici and approximately, 25 different crosses were made with North American and Israel isolates. Progeny of crosses were characterized on standard wheat differentials and increased. Collections derived from these crosses were supplied for genotyping and further analysis. Morphological characterization of spores and nuclear states was completed for several rust fungi including: P. striiformis f. sp tritici, P. coronata, P. graminis, and Uromyces viennot-bourginii. In addition, collections of P. striiformis, P. triticina, P. hordei, P. allii, and U. scillarum were made.
