Title: GENETIC DIVERSITY AMONG 23 USDA INBRED LINES USING CDNA PROBES

Interpretive Summary: Sunflower is a major oilseeds crop and is indigenous to the continental U.S. The narrow genetic background in cultivated sunflower has long been a main concern for breeding programs worldwide. The improvement of interspecific gene transfer from wild species into cultivated lines in recent years will greatly broaden the genetic base of this crop and offer higher selection potential for further improvement. Molecular tools such as cDNA probes are useful for the characterization of genetic diversity, monitoring gene transfer, construction of linkage maps, assisting the mapping of classical genes, and leading toward marker-assisted selection for both major gene and QTL traits. Since USDA released germplasm lines have been widely used by breeders all over the world, we decided to evaluate their genetic diversity before approaching the construction of our public RFLP linkage map for cultivated sunflower. The similarity matrix of the 23 lines showed that, with the exception of 0.99 between HA822 and cmsHA822, the similarity values varied from 0.20 (RHA271-HA372) to 0.85 (RHA274 RHA358). HA822 and cmsHA822 were used as internal checks and the results indicated a nearly identical pattern for all the alleles examined. The 272 alleles were detected by 79 probes averaged to 3.4 alleles per probe. The average similarity index among all the lines was 0.49, excluding that between HA822 and cmsHA822. In general, the majority of the similarities were between 0.40 and 0.60, indicating the good diversity of the representative USDA germplasm lines.
