Title: DELIVERY AND EXPRESSION OF THE GREEN FLUORESCENT PROTEIN (GFP) GENE IN OAT,BARLEY AND MAIZE, AND UTILIZATION AS A SELECTABLE MARKER

Technical Abstract: The objectives of our research were to visually select stable transformants from oat, barley and maize tissue cultures transformed with the gfp (green fluorescent protein gene) and to compare the efficiency of selection with that of traditional herbicide and antibiotic resistance selection schemes. The gfp gene was codelivered with the nptII or bar gene into embryogenic tissue cultures of oat, immature half-embryos of barley, and nonregenerabl suspension culture cells of maize via microprojectile bombardment. Several putative transgenic oat and maize cell lines uniformly expressing gfp, and barley lines displaying chimeric gfp expression have been recovered following visual selection. Herbicide and antibiotic resistant oat, barley and maize cultures have been recovered following growth on selective media. Results from molecular analyses of cell lines and regenerants, and data comparing the efficiencies of selection schemes will be presented.
