Title: INDUCTION OF APOPTOSIS IN MADIN-DARBY BOVINE KIDNEY CELLS BY PSEUDORABIES VIRUS

Technical Abstract: The process of cell death as a result of pseudorabies virus (PRV) infection in Madin-Darby bovine kidney (MDBK) cells was examined. Morphologic changes of the cells after exposure to PRV were monitored by light and electron microscopy. Features characteristic of cells undergoing apoptosis were observed and they included cell shrinkage, chromatin condensation and margination, and apoptotic body formation. Changes in cellular DNA integrity were assayed by cellular DNA fragment ELISA, terminal deoxynucleotidyl transferase-mediated nick-end labeling (TUNEL) assay, and agarose gel electrophoresis. Cellular DNA degradation and fragmentation inside the cells were detected; however, DNA ladder formation was not apparent. Western blot analysis showed that a Bcl-2 related protein was cleaved during PRV infection. These results demonstrated that PRV induces apoptosis in cultured MDBK cells.
