Title: ANTIOXIDANT ACTIVITY OF METHANOL EXTRACTS FROM THE SEED COAT OF VARIOUS PHASEOLUS VULGARIS L. GENOTYPES

Technical Abstract: Natural antioxidants such as flavonoids in foods are receiving attention for their potential beneficial role as free radical scavengers. We sought to identify the flavonoids in the seed coat of P. vulgaris from genetic stocks in which the color determining loci are known. The common names of the stocks are: mineral brown (MB), mat soft brown (MSB), yellow brown (YB), buffy citrine (BC), dark brown violet (DBV), mat black (MTB), grey white (GW), 5-593, V0400 and V0059. Seed costs were removed, lyophilized, ground to a fine powder and then extracted sequentially with hexane, EtOAc and MeOH. Antioxidant activity was tested by fluorescence spectroscopy of liposome oxidation. The hexane extracts showed no activity but the EtOAc extracts from MB, DBV and BC had greater antioxidant activity at 10ppm over a period of 21 min than the MeOH extracts. However, all MeOH extracts from the ten genotypes showed good activity. Two compounds from MeOH extract of one genotype (MB) have been isolated and identified as (+/-) dihydroquercetin and (+/-) dihydrokaemphferol. Further analysis will determine if these compounds are in part responsible for the antioxidant activity of the methanol extracts.
