Title: Flavonol Glycosides from the Seed Coat of Manteca Beans

Technical Abstract: Manteca bean is a variety of Phaseolus vulgaris L. native to South America that has been noted for its ease of digestion and low flatulence. One Kg of Manteca beans was washed and placed in distilled water for 1-2 h and the seed coats removed. Seed coats were then lyophilized, ground to a fine powder and extracted sequentially with hexane, EtOAC, MeOH and MeOH:H2O 1:1. 100 mg of the MeOH crude extract was dissolved in 1.6 ml MeOH and 0.4 ml of H2O added. The extract was then centrifuged at 10,000 RPM for 10 min and the pellet recovered in CHC13. The CHC13 and MeOH:H2O fractions were subjected to TLC analysis with silica gel and C18 plates respectively. Five compounds were present in the MeOH:H2O fraction and 6 compounds in the CHC13 fraction. The MeOH:H2O fraction was further purified by recycling preparative liquid chromatography (LC20). Two isolated compounds were identified as (+/-) dihydrokaempferol-3-O- glycosides by **1HNMR. Further isolation of seedcoat compounds is underway Future work involves identifying all flavonoids Manteca beans and conducting digestibility tests.
