Technical Abstract: Gynogenetic (isogeneic) fish offer enhanced opportunities to study MHC function in teleosts. To this end, members of a putative family of second-generation gynogenetic catfish were characterized by DNA microsatellite, Southern blot, and cytotoxic assay to determine if they were structurally and functionally isogeneic. Microsatellite DNA analysis suggested that all of these fish were closely related. Southern blot analysis, using a catfish MHC class II . gene probe, revealed identical banding patterns among putative gynogenetic fish. In contrast, banding patterns from outbred fish differed not only from gynogenetic animals, but also among themselves. In addition, cytotoxic cell assays demonstrated that peripheral blood leukocytes (PBL) from gynogenetic fish lysed cells from outbred animals, but not those of presumed family members, whereas PBL from outbred fish lysed both gynogenetic and allogeneic targets. Taken together, these results indicate that an isogeneic family of gynogenetic catfish has been developed, and suggest that these fish may provide an excellent system in which to study the role of MHC in immunity.
