Interpretive Summary:

Technical Abstract: Flow cytometry was used to characterize isolates of Phialophora gregata using the fluorescence intensity of PI-stained conidia. Fungal isolates differed in their mean fluorescence intensity, ranging from 116.4 to 129.7 AU's. When the number of fluorescent events were plotted against intensity of fluorescence, a single peak was observed. Fluorescent patterns of Phialophora-like fungi isolated from soybean vascular tissue were compared to isolates of P. gregata. Their mean fluorescence intensity ranged from 76.4 to 88.0 AU's. Multiple peak histograms were observed corresponding to multiple spore sizes as well as single and double nucleated conidia. Genome size, as measured in bp, was estimated by comparing the mean fluorescence of sorghum nuclei to that of the conidia. The relative DNA content of isolates of P. gregata ranged from 8 to 11 x 10 bp, while that of the Phialophora-like isolates ranged from 5 to 7 x 10 bp. Using flow cytometry, we were able to distinguish P. gregata isolates from Phialophora-like fungi isolated from soybean vascular tissue, based on mean fluorescence intensity and/or the presence of multiple peaks
