Technical Abstract: Bovine genome maps are supported by three types of data, i.e., synteny, linkage and cytogenetic information. These sources of information are gradually being integrated into a comprehensive genome map. Integrated maps will be invaluable for the identification of candidate genes once trait loci have been mapped by linkage with polymorphic markers to specific bovine chromosome regions. Cytogenetic anchors are also a prerequisite for using comparative maps to identify candidate genes for trait loci. The positional candidate gene approach is particularly attractive for cloning quantitative trait loci in domestic species, where resources for straight positional cloning are limited. Finally, comprehensive (integrated) maps facilitate microdissection and microcloning of chromosomal regions containing a trait locus. Large genomic clones containing microsatellites have been used for the integration of different information in cattle. Polymorphic microsatellite within large genomic clones allow simultaneous fluorescent in situ hybridization (FISH) and genetic mapping. We report the mapping of a lambda clone to chromosome 27, the last bovine chromosome without an assignment. This assignment anchors the unassigned linkage group B, since the mapped lambda clone contains the BM203 microsatellite.
