Technical Abstract: The northern corn rootworm (NCR, Diabrotica barberi) in eastern and central North America exhibits at least three distinct populations with respect to Wolbachia infection: uninfected; singly infected; multiply infected. The infected states are associated with different mtDNA haplotypes and reduced mt variability. To better describe the multiplicity of infection, Wolbachia gene fragments were cloned from individual NCRs. Gene sequences were obtained from the MLST (Multi Locus Strain Testing) gene group (ftsZ, fbpA, gatB, hcpA, and coxA). For ftsZ two different primer pairs were employed that resulted in partially overlapping fragments. The number of clones sequenced per gene per individual ranged from 1-20. Sequence clusters were identified using Neighbor-Joining analysis. MLST sequencing indicated that the multiple infections include at least five distinct strains. GenBank accessions represent DNA sequences from 240 clones.
