Title: Physical mapping of puroindoline b-2 genes in wheat using the 'Chinese Spring' chromosome group 7 deletion lines

Interpretive Summary: The puroindoline genes (puroindoline a and puroindoline b), located on the distal end of the short arm of chromosome 5D, have a significant effect on grain hardness. Puroindoline b-2 was another puroindoline-like gene after Gsp-1 (Grains Sofftness Protein). The detail position Puroindoline b-2 is still not clear, even though Pinb-2v1, Pinb-2v2, Pinb-2v3 and Pinb-2v4 were mapping on 7AL, 7BL, 7B and 7DL, respectively. The deletion stocks are valuable tools for studies on basic chromosome biology, such as the healing of broken ends, and physical genome mapping. Further research on the physical mapping of Puroindoline b-2 variant genes, using 24 homozygous deletion lines of Chinese Spring, found that Pinb-2v1, Pinb-2v2 and Pinb-2v4 were located on the distal end of the long arm of chromosomes 7D, 7B and 7A, respectively.

The detail position Puroindoline b-2 is still not clear, even though Pinb-2v1, Pinb-2v2, Pinb-2v3 and Pinb-2v4 were mapping on chromosome 7AL, 7BL, 7B and 7DL, respectively. The deletion stocks are valuable tools for studies on physical genome mapping. In this study, Puroindoline b-2 variants were physical mapping using 24 Chinese Spring (CS) of group 7 deletion lines, including 7 deletion lines of chromosome 7AL (CS Del7AL), 14 deletion lines of chromosome 7BL (CS Del7BL) and 10 deletion lines of chromosome 7DL (CS Del7DL). Results indicated that Pinb-2v1, Pinb-2v2 and Pinb-2v4 were located on the distal end of the long arm of chromosomes 7DL, 7BL and 7AL, respectively. The study of physical mapping of Puroindoline b-2 in bread wheat would contribute to further understand the molecular and genetic basis of kernel hardness.
