Interpretive Summary:

Technical Abstract: Recent studies of the sirtuin family of proteins, which possess NAD+/-dependent deacetylase and ADP ribosyltransferase activities, indicate that they regulate many biological functions, such as longevity and metabolism. These findings also suggest that sirtuins might serve as valuable drug targets for the treatment of a variety of diseases, ranging from diabetes, cardiovascular diseases, neurodegenerative disorders and cancer. This chapter reviews the current knowledge of sirtuins' functions in metabolic regulation and their emerging roles as therapeutic targets for the metabolic syndrome.
