Technical Abstract: NADH dehydrogenase, the largest of the respiratory complexes, is the first enzyme of the mitochondrial electron transport chain. We have cloned and sequenced cDNA of NADH dehydrogenase gene from Ochlerotatus (Ochlerotatus) taeniorhynchus (Wiedemann) adult (GeneBank Accession number: FJ458415). The full-length mRNA sequence (824 bp) of NADH dehydrogenase Oc. taeniorhynchus (OctNADH) encodes an open reading region of 651 bp (i.e., 217 aa). To detect whether OctNADH dehydrogenase of the mosquito Oc. taeniorhynchus is developmentally regulated, we used quantitative real-time polymerase chain reaction (qPCR) to examine NADH dehydrogenase gene expression levels in different developmental stages of Oc. taeniorhynchus. qPCR showed that OctNADH was expressed in each developmental stage and highly expressed in the teneral adult stage. qPCR also showed that mRNA transcription levels of OctNADH were up- or down-regulated during the time course study in response to permethrin treatment in female Oc. taeniorhynchus. In addition, OctNADH5 dehydrogenase, mitochondrially encoded NADH dehydrogenase subunit 5 (GeneBank Accession number: FM992318), was also examined and compared two different subunits of Oc. taeniorhynchus.
