Technical Abstract: In order to preserve and develop genetic resources important to the apple industry, the USDA-ARS Plant Genetic Resources Unit has established a globally representative collection of Malus. Malus sieversii, the main progenitor of the domestic apple collected from Central Asia (Khazakhstan), is among this collection. In order to identify genomic regions associated with important traits in M. sieversii, we have undertaken the construction of a genetic framework map for the family (F1) GMAL4593 ('Royal Gala' X M. sieversii PI 631981 [GMAL 4448]). GMAL4593 was subjected to a combination of SSRs and SNPs analyses. Approximately 400 molecular markers were screened in the entire population (199 individuals) or in a population subset (96 select individuals). Over 60 percent of the markers were polymorphic in the population and were used for the construction of the genetic framework map. The map was constructed using Joinmap 3.0 for an outbreeder full-sib family population type. Currently, the 'Royal Gala' map consists of 163 molecular markers assigned to 16 linkage groups, and the Malus sieversii map consists of 75 molecular markers assigned to 14 linkage groups. We are in the process of screening additional SSR and SNP markers in order to complete a genetic framework map that is representative of all 17 Malus linkage groups. Initial phenotyping of the family for fire blight resistance has been conducted, and a preliminary QTL analysis of the data will be presented.
