Technical Abstract: Zucchini yellow mosaic virus Florida stain (ZYMV-FL) is one of the most economically important viruses affecting watermelon in the United States. Inheritance of resistance to ZYMV-FL is conferred by a single recessive gene. Described here is single-reaction, polymerase chain reaction-based marker linked to the ZYMV-FL resistance gene in watermelon. This marker, ZYRP, was found to be linked to the ZYMV-FL resistance gene locus (genetic distance of 8 cM) in a F2 population, as well as in a population derived from a backcross of an F1 to the resistant parent (BC1R) (genetic distance of 13 cM). The identification of a single nucleotide polymorphism within the ZYRP marker for the parental genotypes allowed the development of a sequence characterized amplification region (SCAR) marker linked to the ZYMV-FL resistance gene. The ZYRP marker also was found to be useful in a marker assisted selection strategy to identify the introgression of the gene conferring ZYMV-FL resistance to a susceptible watermelon cultivar from a BC2F2 population derived from the PI 595203 (ZYMV-FL resistant) and the recurrent parent ‘Charleston Gray’ (susceptible).
