Technical Abstract: Research on methods for molecular-genetic testing of the Septoria pathogens of wheat was initiated. Two species of septoria were studied: Septoria tritici Rob. et Desm., and Stagonospora nodorum [Berk] Castellani and E.G. Germano. Different protocols for extraction of DNA were applied; the best method of DNA extraction depended on the quantity of the pathogen. Quantitative and qualitative characteristics of DNA samples were investigated. The work on optimization of PCR conditions with ITS primers was carried out: the reaction components, its temperature and time indicators were defined. As a result, the research revealed that Stagonospora nodorum and Septoria tritici show a positive reaction with primers designed to specifically amplify DNA from these phytopathogens. The conditions developed for detection and identification of the two Septoria species - Stagonospora nodorum and Septoria tritici - with defined molecular markers will allow future research on identification of these pathogens to be conducted in vivo directly in seeds and in the plant.
