Technical Abstract: In 2006 and 2007, over hundred potato varieties were screened for potato tuberworm Phthorimaea operculella Zeller resistance under field and laboratory conditions. At harvest, potato tubers were graded to select approximately similar size (6-8 oz) tubers. Tubers harvested were stored at room temperature for approximately 10 days to check for tuberworm infestation. Uninfested tubers were stored in cold storage at 4 °C until the screening test started. In the field, two tubers of each clone per rep were placed on the surface of the soil and were exposed to natural populations of potato tuberworm using a randomized complete block design with ten replications. In the laboratory, two tubers were placed in plastic containers and artificially infested with 50 tuberworm eggs laid on filter papers which were placed on top of tubers. Tuberworm damage was recorded three weeks after inoculation by evaluating larval survival, number of mines, and incidence of damage and average of severity. Resistance of potato varieties was measured based on larval survival and damage.
