Technical Abstract: Virus-like symptoms including a mosaic of light and dark green, faint chlorotic spots and rugosity were observed on the leaves of several brugmansia plants in a south Florida nursery in November 2003. The presence of a potyvirus was indicated by inclusion body morphology and electron microscopy of leaf dips. Additional evidence was obtained by testing with a commercially-available enzyme-linked immunosorbent assay (ELISA) for multiple potyvirus species. Sequence analysis of an ~1.7 kb 3’ terminal genome fragment amplified by reverse transcription-polymerase chain reaction from total RNA using degenerate potyvirus primers confirmed the diagnosis. The nucleotide sequence of the amplified product was 99% identical to the corresponding region of Colombian datura virus (CDV) sequences in GenBank. A broader survey of viruses in brugmansia was initiated in subsequent years with samples collected from states in the northeastern, midwestern and western U.S. All samples were assayed for potyviruses (and specifically CDV) and also for tobamoviruses, tospoviruses and Cucumber mosaic virus by ELISA.
