Interpretive Summary:

Technical Abstract: Genomic research can provide new tools and resources to revolutionarily enhance crop genetic improvement and production. However, genome research in peanut is far behind those in other crops, such as maize, soybean, wheat, and sorghum due to the shortage of essential genome infrastructure, tools, and resources. Cultivated peanut is an allotetraploid (2n=4x=40). Because of the large genome and polyploidy, one of many challenges peanut crop faces is the improvement using genetic and genomic approaches. Recently, six cDNA libraries from 3 seed stages (R5, R6 and R7) of two cultivars, Tifrunner (resistant to leaf spots and tomato spotted wilt virus) and GT-C20 (resistant to Aspergillus infection, bacterial wilt, and leaf rust) have been constructed, and 24,096 randomly picked clones have been sequenced from the 5' ends. One full-length cDNA, encoding a new peanut seed storage protein as a potential allergen iso-Ara h3, has been cloned by contig. The deduced amino acid sequence has three of four IgE binding sites of Ara h3. Further Western blot analysis supports that the isoforms of this storage protein on 2-D gel can be detected by Ara h3 antibody. These ESTs will facilitate gene discovery, marker development and gene functional characterization.
