Technical Abstract: Xylan, a major component of hemicellulose, requires a complex of enzymes for complete hydrolysis of the main and side chains. XynA is a bifunctional enzyme with endo-b1,4-xylanase and acetylxylan esterase activities. It has the potential to be an important component of a hemicellulose-degrading enzyme cocktail, taking advantage of the synergism demonstrated between the two active domains. The Clostridium cellulovorans xynA gene was cloned into a yeast expression vector and transformed into Saccharomyces cerevisiae. The enzyme was constitutively expressed and secreted in an active form. The XynA activity was found mostly in the culture medium with residual activity in the cell extracts. Partial purification of the enzyme was achieved by cation-exchange and gel filtration chromatography.
