Title: GENETIC TRANSFORMATION OF SCLEROTINIA SCLEROTIORUM THROUGH AGROBACTERIUM TUMEFACIENS-MEDIATED TRANSFORMATION.

Technical Abstract: In order to study genetic factors of pathogenicity, insertional mutants of Sclerotinia sclerotiorum were generated using Agrobacterium tumefacience-mediated transformation with both mycelial fragments and ascospores. Transformants were tested for number of insertions by Southern hybridization and for stability of hygromycin B resistance, and are being screened for altered pathogenicity on lentil plants and for production of oxalic acid. The tagged mutations will allow identification of mutated genes.
