Technical Abstract: Toll-like receptors (TLRs) is a family of transmembrane proteins able to recognize conserved pathogen structures and induce specific adaptive immune responses. We identified a number of rainbow trout EST sequences with high homology to TLR genes from other species using in silico analysis. The cDNA library inserts were sequenced and used as probes to screen a rainbow trout BAC library (bacterial artificial chromosomes) and identify clones containing homologous sequences to TLR genes. DNA fingerprint analysis results suggest that some TLR genes are present in more than one copy in the rainbow trout genome. The positive BAC clones were sequenced to characterize gene structure and for future promoter analysis and genetic markers identification, which will be integrated into the NCCCWA rainbow trout genetic linkage map. Tissue expression profile was determined in adult rainbow trout through reverse transcriptase PCR. There is evidence that at least six TLR genes are expressed in rainbow trout. Functional analysis of the TLR genes will provide a better understanding of host-pathogen interactions and the genetic basis of disease resistance in fish, for the implementation of more effective health management practices in aquaculture.
