Interpretive Summary:

Technical Abstract: During cereal grain germination, GA is produced in the embryo and then moved to aleurone tissues, where it induces production of many hydrolytic enzymes such as alpha-amylase. Isolated aleurone tissues do not produce endogenous GA itself, but have a strong response to exogenous GA treatment. Thus, isolated barley aleurone tissues provide an excellent system to study GA signal transduction pathways. We examined the GA induced production of alpha-amylase in response to a battery of chemical signals. Using barley genome GeneChips, we surveyed expression of over 22,000 genes in response to GA and a proteasome inhibitor treatments to dissect genomic regulation of GA signal transduction pathways. Examination of the isolated aleurone's reponse to other treatments and the study of GA mutants are in progress. Meanwhile, we cloned barley genes encoding two different F-box proteins, an alpha subunit of G proteins, and their corresponding double strand RNA interference constructs were made. The endogenous gene activities in isolated barley aleurone tissues will be silenced by transient expression of their dsRNAi constructs, and the functionality of the genes in GA signal transduction will be determined by measuring alpha amylase promoter activity in response to GA treatment in the aleurone tissues. The detailed data from the experiments will be discussed.
