Some affected galls contained bacteria only and some contained both bacteria and dormant nematodes. The bacterium was readily cultured and identified phenotypically as a Rathayibacter sp. The bacterium was Gram-positive, non-acid fast, catalase positive, hydrolyzed esculin, failed to produce acid from sorbitol, and oxidase negative. Colonies were round, non-mucoid, yellow, and 1-2 mm in diameter after 7-8 days at 25C on NBY agar. Bacteria from galls and cultures adhered to A. woodi juveniles but not those of A. funesta or A. tritici. As with R. toxicus, the agent of ryegrass toxicity, bioassays indicated in planta production of toxin. DNA from the bacterium had only 70% homology with other Rathayibacter species and gave a distinct AFLP pattern. These data are indicative of a new Rathayibacter species. |
