Interpretive Summary:

Technical Abstract: With the completion of the rice genome sequencing project in December 2004, the next cereal genome to be sequenced will be maize. The maize genome (~2000 Mb) is nearly five times bigger than that of rice (~400 Mb) and is rich in nested retrotransposons, which comprise approximately 80 percent of the genome. Due to genome size and repetitive DNA content scientists are now performing pilot projects to determine the most efficient strategy to sequence the maize genome. Regardless of the approach, a complete sequence-ready physical map of the maize genome will be required. Here, we report our finished map of maize genome. A total of 292,039 BACs, equal to 20x genome coverage, have been fingerprinted on high resolution agarose gels using three BAC libraries (HindIII, EcoRI and MboI). These BAC fingerprints were assembled with FPC and anchored to the genetic map. Hybridization of the BAC libraries with 2000 genetic markers and 16,000 overgo markers, derived from a EST unigene set, and manual editing resulted in nearly 2800 contigs with ~55 percent of the genome anchored. The extensive overgo marker hybridization data allow us to construct a maize expression map as well as a duplication map of the maize genome, providing a strong evidence of tetraploid origin of the maize genome. Here, we will report our integrated physical and genetic map, a transcriptional expression map, a maize genome duplication map, and a maize-rice synteny map.
