Interpretive Summary:

Technical Abstract: Historically, stem and leaf rust have been among the most devastating diseases of wheat, with written accounts of epidemics dating back to ancient Greek and Roman times. In North America, major epidemics have occurred from the late 1800's through the 1970's. During the cold war, the United States and the Soviet Union studied Puccinia graminis f.sp. tritici as a biological warfare agent. Based on current concern for bio-security of agricultural crops, we are developing real-time PCR assays for the rapid detection of P. graminis, P. recondita, P. triticina and P. striiformis using a TaqMan assay system. Using rust specific primers and species-specific FAM probes in the ITS 1 region, we have developed assays for P. graminis and P. striiformis. Current results on the development of real-time PCR assays for these wheat pathogens will be presented.
