Title: LC-MS ANALYSIS OF METABOLITES OF TANGERETIN IN HAMSTER

Technical Abstract: The citrus polymethoxylated flavones have attracted attention as potential health-promoting compounds with anticancer, anti-inflammatory, and cholesterol-lowering properties. While many in vitro studies have substantiated activities coupled to these pharmacological properties, the study of the metabolism and tissue accumulation of these compounds in animal feeding trials are critical to understanding the potential roles of these compounds. In a feeding trial with hamsters, numerous metabolites of tangeretin were detected in the serum, urine and liver extracts by LC-MS. The metabolites in the serum and urine samples consisted mainly of glucuronides, with minor aglycones also present. In contrast, LC-MS analysis of the liver extracts showed the preponderance of aglycones over the glucuronides. Trace levels of intact tangeretin were also detected in the liver. Total levels of metabolites in the liver extracts ranged from 37 ppm to 167 ppm. High levels were also detected in the urine and serum. These results show that elevated levels of tangeretin metabolites occur following uptake through the diet. These high levels suggest that sufficient levels may occur to allow tangeretin to exert biological activities in vivo.
