Interpretive Summary: As a first step in chickpea genome characterization, We have constructed a genomic library which can be used to isolate desired genes, in particluar, disease resistance genes. We have also shown the utilisation of the library to isolate the clones which probably have fusarium wilt resistance gene. This indicates the potential use of the library to isolate disease resistance genes which is one of the major applications. This work is the first step towards chickpea genomics.
Technical Abstract: Bacterial Artificial Chromsome (BAC) library is most commonly used genomic resource for genome characterization in other crops but was absent in chickpea. We identified two BAC clones using a molecular marker linked to fusarium wilt resistance gene to show the use of BAC library in map based cloning. We characterized these two clones by checking the genomic insert size and by sequencing at both ends. Also we compared the end sequences of our BAC clones with Medicago which is a model legume. This essential tool is available for all chickpea researchers.
