Interpretive Summary: A new method for the study of whole sections of rice kernels was developed. The new method will allow scientists to quickly determine the relationships between kernel cellular components, how the components differ between cultivars, and how the components change during or as a result of processing. The work is expected to expand on the definition and determination of quality attributes in rice.

Technical Abstract: A new and simple method was developed to collect sections of a whole brown rice kernel for investigation of histological properties. A single kernel of rice was dehydrated through a graded ethanol series, transferred to xylene, and embedded in paraffin. During sectioning of the blocks using a rotary microtome, a special adhesive tape was used to collect and place the sections onto slides so that they remained flat. The use of the adhesive tape technique combined with autofluorescence characteristics allowed the visualization of cell walls throughout an entire longitudinal or transverse section of a whole grain or milled rice kernel. SEM images of the sections were also obtained.
