Interpretive Summary:

Technical Abstract: Leaf rusts caused by Puccinia recondita sensu lato and P. hordei s.l. are major diseases of cereals and grasses worldwide. Uredinologists have used two concepts for taxonomy of the above rusts viz., broad or narrow. We obtained nucleotide sequences of the rDNA region (ITS1, 5.8S rRNA gene, and ITS2) from dried herbarium materials (up to 20 years old) of leaf rusts on various hosts from Iran to determine their phylogenetic relationships. Our results revealed that both ITS1 and ITS2 contained phylogenetically informative sites. Molecular analysis carried out in the present study agreed with the narrow-species concept for graminicolous rusts. The cladistic analysis of ITS data suggests recognizing P. holcina (on Lolium and Lophochloa) as a separate species from P. hordei (on barley). Moreover, ITS data verified P. recondita (on Secale), P. bromina (on Bromus), P. persistens ssp. triticina (on wheat) and P. persistens ssp. agropyrina (on Elymus) as authentic taxa. We suggest using P. persistens ssp. triticina for brown rust on wheat rather than P. triticina.
