Technical Abstract: The initial purpose of this pioneering study was to determine if AFLP markers could be used to determine the amount of genetic variation in Trifolium subterraneum and its usefulness in discovering duplications in the collection and potential gaps of genetic diversity. One hundred and twenty-five accessions from the subterranean clover collection were assayed by AFLP analysis. A total of 633 bands from three different primer combinations generated a UPGMA tree with a high degree of robustness when tested using mantel statistics. Each primer set generates a different banding pattern allowing for greater genome coverage. As the clover accessions are field increased, phenotypic descriptors are observed and compared to the AFLP and geographical data. When the entire study is completed, over 250 accessions of subterranean clover will be assayed with about 5,780 data points for the phenotypic descriptors and over 900,000 data points for the AFLP markers. Within, between, and among variation using the AFLP data will be used to access the genetic diversity in the entire collection of subterranean clover.
