Technical Abstract: Acid (ACP) and alkaline phosphatase (ALP) activities were examined in relation to adult development and diapause in both soluble and membrane- bound extracts of the midgut, hindgut and Malpighian tubules in the Colorado potato beetle, Leptinotarsa decemlineata (Say). The soluble ACP activity was higher than the membrane-bound ACP from all tissues examined, with different developmental profiles among tissues between long- and short-day beetles. The highest soluble ACP activity was found in the midgut contents, while a maximum soluble ALP activity showed in the Malpighian tubules. Treating short-day diapausing beetles with JHA or allatotropin had significantly inducive effect on both ACP and ALP activities in all tissues examined. Treatment with 20-HE alone did not increase ACP activity in any tissue, but did increase ALP activities in all tissues except the midgut. Furthermore, JHA was able to enhance and/or modulate the effect of 20-HE treatment.
