Technical Abstract: The horn fly, Haematobia irritans, is an obligate hematophageous parasite of cattle causing significant economic losses to cattle producers. The insect overwinters at the pupal stage in a state of facultative diapause, the onset of which seems to be primarily induced by decreasing temperature. In order to begin to understand the molecular regulation of the induction of diapause in the horn fly, we have utilized the technique of differentia display PCR to compare gene expression between horn flies developing in diapause-inducing conditions and flies developing in diapause-averting conditions. Larval and early pupal stage samples reared at 15oC (diapause-inducing) were compared to samples reared at 25oC (diapause-averting)) and RT-PCR products from differentially expressed mRNAs were cloned and sequenced.
