Technical Abstract: This book chapter briefly reviews previous research with peanut in the area of genetic transformation, including Agrobacterium-mediated gene transfer techniques. Detailed and previously unreported protocols are presented for the regeneration of cultivated peanut from isolated mesophyll cell protoplasts and for the transformation of these protoplasts with plasmid vectors containing marker genes or gene expression cassettes containing various virus coat protein genes. The chapter also contains the results of a series of experiments that resulted in the successful transformation of peanut with a strain of Agrobacterium tumefaciens harboring a vector containing a gene expression cassette that included the nucelocapsid protein from an isolate of the tomato spotted wilt virus.
