Title: Certain Subunits of Plastid Acetyl-Coa Carboxylase from Soybean Are Encoded by Multigene Families

Technical Abstract: Acetyl-CoA carboxylase (ACCase) carries out the primary reaction in de novo fatty acid biosynthesis. Our research has focused on the multisubunit form of the enzyme that resides in soybean chloroplasts. A clear understanding of the mechanisms that control the activity of this ACCase in plants is lacking. It is believed to be the site where carbon flux into the FAS pathway in plants is regulated. We investigated the organization of genes representing the multimember families of MS ACCase subunits in soybean. Genomic DNA that corresponded to four alpha-CT genes, two BC genes and a BCCP gene were sequenced and analyzed. Gene expression profiles were performed for each of the four ACCase subunits. Major differences in the steady-state mRNA levels among family members were evident at different time points during soybean development. The BC-2 gene produced fairly constant amounts of mRNA throughout seed development and germination, whereas BC-3 exhibited a remarkable mRNA burst early during the formation of seeds. Alternative splicing of mRNA for an alpha-CT may also occur.
