Title: MOLECULAR CYTOGENTIC CHARACTERIZATION OF BROMEGRASS

Technical Abstract: Previous cytogenetic studies of Bromus inermis and related Bromus species were limited to chromosome counts and construction of karyotypes based on feulgen staining. Since the chromosomes of the polyploid bromegrasses are similar in morphology, these karyotypes were of limited use for genome analyses. We used molecular cytogenetic markers to develop karyotypes for three Bromus species. We standardized a C-banding procedure for bromegrasses. We have made a karyotype of diploid (Bromus riparius), tetraploid (Bromus inermis) and octaploid (Bromus inermis) based on C- banding patterns, centromeric location and chromosome size. All chromosomes of the diploids, tetraploids and octaploids had clearly visible C-bands except two pairs of octaploid chromosomes. The C-banding pattern of the diploid Bromus riparius is quite different from that of tetraploids. Therefore, diploid B. riparius could be eliminated as a possible progenitor rof the tetraploid Bromus inermis. Preliminary results suggest that octaploid B. inermis may not be a doubled form of the tetraploid B. inermis due to differences in C-banding patterns of chromosomes.
