Title: LINKAGE MAPPING OF TWO TAQ1 PCR-RFLPS WITHIN INTRON 6 OF THE PORCINE GENE (LCN1) ENCODING TEAR LIPOCALIN/VON EBNER'S GLAND PROTEIN

Interpretive Summary: The gene encoding the Tear Lipocalin Gland Protein (LCN1) was mapped to the end of the long arm of chromosome 1 in the pig. The assignment of this gene increases the number of genes assigned to this porcine chromosome and supports the development of the comparative map between pigs and humans in this region.

Technical Abstract: Source/description: Primers pTP6s and pTP7a designed by Holzfeind et al. based on the cDNA sequence were used. On porcine genomic DNA sequence forward pTP6s and reverse pTP7a primers match at positions 3326-3346 and 4380-4400, respectively, giving a 1064 bp PCR product encompassing parts of exons 6 and 7 as well as all of intron 6. Polymorphism: Two polymorphic restriction sites (RES) were found within the PCR fragment. They are at positions 3373 and 3780 with corresponding loci TaqI 3373 and TaqI 3780, respectively. Mendelian inheritance/allele frequencies: Codominant Mendelian inheritance of alleles in both loci was demonstrated. Allele frequencies in eight breeds are given in Table 1. Allele TaqI3373 B was fixed in all breeds with exception of Minzhu. Allele TaqI3780 A was abundant in all breeds under investigation while allele TaqI3780 B was found in Pietrain, Landrace and Czech Meat Pig. Apart from these breeds also individual animals of Duroc (n=2), Fengjing (n=1), and European Wild Boar (n=1) were typed. In all of these animals only alleles TaqI3373 B and TaqI3780 A were detected. Linkage mapping and chromosomal localization: Multipoint linkage analysis was performed in USDA-MARC backcross pedigree using CRIMAP software package, version 2.4. The gene has been located on USDA-MARC 2 linkage map to SSC1 at position 140.8 Kosambi cM in following order: SW1301-(0.3)-LCN1-(2.3)-SW2513.
