Technical Abstract: RAPD and AFLP markers linked to the genes B (Beta) and hp (high pigment), two of the genes involved in the production of carotenoids in tomato, were identified using near isogenic lines (NILs) and bulked segregant analysis of an interspecific F-2 population derived from the cross of Lycopersicon cheesmanii accession LA 371 x L. esculentum cv. Floradade. A total of 600 10-mer RAPD primers and 64 AFLP primer combinations were screened in this study. A number of primers (RAPD) and primer combinations (AFLP) detected polymorphism between bulks from the interspecific F-2 population or between the pairs of NILs. F-2 and BC populations derived from B iso-Rutgers x Rutgers and hp iso-Rutgers x Rutgers were used to confirm marker linkage. A single RAPD product was identified which cosegregated with the hp phenotype in the F-2 population from hp iso-Rutgers x Rutgers. Similarly, a single AFLP primer combination was identified which detected a polymorphic product cosegregating with the B phenotype in the F-2 population from B iso-Rutgers x Rutgers. Additional markers detecting polymorphism between the respective bulks or pairs of NILs are being evaluated for potential linkage with the B or hp phenotype.
