Technical Abstract: A population of horn flies was found in Central Texas which exhibitied significant resistance to pyrethroid insecticides and the OP diazinon as determined by filter paper bioassays. A PCR-based subtractive hybridization technique followed by RNA Northern blot analysis was utilized to examine differences in gene expression of this resistant field population compared to a susceptible laboratory colony. To determine if a mutated esterase gene might be involved in the development of diazinon resistance in this population, RT-PCR using several degenerate primers designed from conserved regions of other Dipteran esterases involved in OP resistance were used to clone members of the horn fly esterase gene family. The DNA sequences and open reading frames of esterase genes from this resistant population were compared to those from the diazinon susceptible laboratory colony.
